The function of catecholase Essay Example for Free

The function of catecholase Essay 5 mL of enzyme, and all of the trials were set up as two tubes to begin with. One tube had 3. 5 mL of water and 2. 0 mL of catechol, and the other had . 5 mL of enzyme. By keeping the enzyme and substrate separate, we were able to have more control over the starting point of the reaction, and were able to bring each individual component to the desired temperature before starting. The temperatures that our group tested were 37oC, 45oC, 50oC, 55oC, 65oC, and 75oC. Both our group and group A-4 tested 37 and 45 degrees. This allowed us to compare each groups results more accurately. The setup for the actual experiment was that each temperature had its own test tube rack. Four tubes of water and catechol and four tubes of enzyme were put into each rack, and these racks were then submerged into either a hot water bath or a circulating water bath, depending on which temperature was being tested. The tubes were allowed to sit in the water for approximately five minutes to allow the contents of the tubes to come up to the temperature we were trying to test. Then the tube of enzyme was poured into the tube of catechol, the tube was covered and inverted to mix the contents, and the timer was started. We allowed the reaction to run from three to five minutes, keeping track of exactly how long each set went for. Our method for ensuring that the reactions ran for the exact length of time stated was to start each reaction thirty seconds later than the first, and then take the measurements from the spectrophotometer exactly thirty seconds apart, keeping the tubes in the same order. Before measuring each tube, we again inverted it several times to make sure that the product was uniformly distributed throughout the tube. Results Results for Group A-5 Temperature (degrees Celsius) Trial 1. Trial 2 Trial 3 Trial 4 Mean Standard Deviation 3 (Table 2) (Graph 1 (Graph 2)According to our results, the optimal temperature for the reaction of catecholase is 45oC. We were able to determine this because the average reaction rate was highest at this temperature (see table 1). We were correct in assuming that the reaction would gradually speed up as the temperature increased until it reached a point where the enzyme began to denature, and then the reaction rate would slow after that. This gradual increase, peak, and then decrease can be seen in the graph 1 above. The bars above and below each data point are representations of the standard deviation associated with each value. This shows how much variation we had within each set of trials. When we compared our results to group A-4, it was clear that they too found the peak reaction rate to be at the 45-degree temperature. This served to further verify our results. It is interesting to note that in the graph of both of our data, although the actual values are not the same, the overall conclusions are. Discussion After analysis of the data, it is clear that our hypothesis was correct, because the found peak does fall within the range of 23 and 75. We can be fairly certain that our data are correct because we were able to control the fixed variables, such as the amounts of each component of the reaction, very well by having the same person measure out the same thing for each trial. We were fairly accurate at controlling the temperature because the baths were monitored at a steady temperature for the time the reaction was taking place, although it would have probably been to our benefit to actually take the temperature of the liquids inside the test tubes instead of only monitoring the environment outside of the tubes. Also, we could have increased accuracy by having the spectrophotometer nearer to the baths, because there was some heat lost during transport, which could have affected the tubes that were measured later. There was also some confusion at the beginning of the experiment about exactly what technique was to be used, which resulted in our having to repeat the trials for the 65-degree temperature. We assume that similar problems are to blame for the differences in the reaction rates for 37o and 45o between our group and group A-4. We also discovered after the experiment had been completed that the other group had left the catechol out in a separate tube during heating, while we left the enzyme out. This difference in method may have contributed to the differences. To make sure that these problems did not result in faulty conclusions, I looked up another experiment on the effects of temperature done by Pierre Greenway. Greenways findings indicate that the peak temperature for enzyme function is actually at 40oC, and not 45. This is an interesting discovery, since we did not test thoroughly in the range of 37o-45o. The next course of experimentation suggested by these findings would be to thoroughly test the reaction rates at temperatures between 35 and 45 degrees Celsius. Measuring at one-degree intervals would give the best results, but may require another collaboration to be achieved in a reasonable amount of time. It may also be interesting to test the reaction rate at even higher temperatures, and try to find the point at which all of the enzyme is completely destroyed. According to the results of Greenway, this temperature was around 60oC for him, but we tested beyond that in our own experiment and found the rate to still be decreasing. Any of these questions would be interesting to try to answer.   Show   The above preview is unformatted text This student written piece of work is one of many that can be found in our GCSE Patterns of Behaviour section.

Enterprise Rent A Car Recruitment and Selection

Enterprise Rent A Car Recruitment and Selection In present business world, organization always tries to reduce their cost and maximize their profit. In this point of view Enterprise rent A-car are not different, they always try to use modern technique in their recruitment, it was founded in 1957 by jack Taylor in St. Luis in the USA. At present it is the largest car rental business in the world today with more than 7500 offices in the USA and more than 850 in Canada, Puerto Rico, the UK , Germany and Ireland. In 2007, organization had more than 700000 rental cars in use, employing over 60000 people with an annual turnover of over  £ 4.6 billion Human Resource Management at Enterprise Rent-A-Car: Human Resource Management does the hiring, training, and developing staffs and where necessary to discipline or dismiss them. Enterprise Rent-A-Car gives extensive training and development program to its employees within the company. This reduces the need for external recruitment and makes maximum use of existing talent. This is a cost-effective way for a business to manage its people. The HRM function not only manages existing staff, it also plans for changes that will affect its future staffing needs. This is known as workforce planning. For example: †¢ the business may grow into new markets, such as Enterprise moving into truck rental †¢ it may use new technology which requires new skills e.g. global positioning equipment †¢ staff may retire or be promoted, leaving gaps which need to be filled. There may also be external changes in the labour market, meaning that there will be fewer skills available or too many in a particular area. HRM monitors all of these things in planning recruitment strategy. This places the HRM function in a central role in the business because all managers use this expertise to acquire staff. Demand and Supply forecast: Every year Enterprise Rent-A-Car requires a higher volume of employee recruitment. Over 65000 people are currently working in Enterprise. With the diversification of the company in Canada, Puerto Rico, the UK, Germany and Ireland, there is a higher demand of the work force in Enterprise all the time. So, the company continuously look for the talented people to recruit. For that reason each year, Enterprise recruits an average of 1,000 staff into its graduate recruitment programme in the UK and Ireland. Recruitment Plan at Enterprise Rent-A-Car: Enterprise has a policy of promoting its managers from within its existing workforce. This means the business must recruit people with the potential to grow. Each year, Enterprise recruits an average of 1,000 staff into its graduate recruitment programme in the UK and Ireland. To achieve its aims and objectives, Enterprise needs staff who are motivated and who possess initiative and drive. Promoting managers and offering career opportunities from within the company has a positive affect on Enterprise. Employees remain happy, will stay longer and give their best. However, with growth and diversification there is always a need for external recruitment to provide new skills or increase the business capacity for expansion. Recruitment and Selection Method: In order to attract high quality candidates, Enterprise is raising the company profile within UK universities using Campus Brand Managers. These are students or interns who work for Enterprise and act as liaisons for potential applicants. Students can also visit Enterprise and spend time learning about how it does business and what opportunities it offers. Enterprises online recruitment process is an important part of its strategy. This improves the speed and efficiency of the application for both the company and the applicant. The website provides a registration function and lists available jobs. It also provides a lot of information about the Management Trainee role and the company culture and values. This allows applicants to get a good idea of whether Enterprise would suit them. Enterprise advertises its vacancies and opportunities across a wide range of media. This includes media such as newspapers, magazines and online. Selection is the process of identifying the best candidate for the role in question. This is important as the candidates who apply may not always have the correct set of skills and competencies required by the business. Enterprise seeks competencies in its recruits both for an immediate job role and also for development over the longer term to support the business growth. HR managers often use standard documentation in order to match job roles with personal qualities and skills. These include: †¢ the job description this summarises a job role within an organisation and lists the main tasks †¢ a person specification this highlights the characteristics a candidate needs for a post, as well as the desirable qualities the company is looking for. Enterprise combines the person specification within the job description by using a skills and competencies framework. Interview Panels and Types of Interviews: The Enterprise rent-car always tries to create best opportunity for their employee. †¢ To screen candidates, Enterprise recruitment managers compare the online application forms (which reflect candidates CVs) to the skills and competencies the role needs. †¢ Candidates then have an initial face-to-face interview with an Enterprise recruitment manager. †¢ This is followed by an interview with a branch manager. †¢ From this, selected candidates are invited to an assessment day. The assessment day is a standard part of the Enterprise recruitment process. Candidates take part in practical exercises, including role-play, as well as individual and group activities. Role-play is a valuable way of testing core skills like communication and customer service. Enterprise can assess a candidates performance by different methods and in different work related tasks. This makes the selection process fairer. Areas tested include customer service skills, flexibility, sales aptitude, work ethic, leadership and teamwork. The assessment day ends with another interview with a senior manager in order to make the final selection. Conclusion: In conclusion we can conclude that, human resource recruitment plan is very effective decision for any successful organization. Organization Recruitment plan are based on the organization demand and responsibility. Enterprise recruitment plan are always different than other similar organization, they always focus on young generation because their main goal is to provide best service for their customer.

Contrast Essay Between essays Just Take Away Thier Guns and I Want A Wi

"Just Take Away Their Guns" is an effective argument than "I Want a Wife." James Wilson gives a more factual, organizational and better language used argument than Judy Brady. In "I Want a Wife" Brady is stereotypical of what she believes to be the roles of the husband and wife. Also Brady's essay is one sided, she never states a side that would argue her viewpoint. While Wilson shows two clearly stated sides of his argument. The first reason as to why ?Just Take Away Their Guns? is more effective is because the essay has more specifics and facts that can?t be refutable. The president wants more gun control but the public says the laws will fail who is right? And in deciding who is right will any of our amendments be taken away or shortened? Taking away the right to be able to buy and own a gun will have hardly any effect on the illegal use of guns. Wilson states that there are 200 million guns in private ownership, about one-third of them handguns. Of the 200 million handguns that are owned, only 2 percent of the population is allowed to commit a crime. Out of the 200 million guns, about one-sixth of them, are bought legally through either a gun or pawn shop. Many of the current handguns owned, are stolen, borrowed, or obtained through private purchases that wouldn?t be affected by gun laws states Wilson. If there was a successful effort to take away or to shrink the amount of legally purchased w eapons or accessories (ammunition) then what happened to our right to bear arms? Isn?t self defense a key part in our time? If so many guns are stolen, then how is taking away the legal guns going to help? It is not, because people will always find ways to get a gun no matter the restrictions set upon it. Although Wilso... ...e of her duties is to care for the kids, and to obey the husband?s wishes of privacy and sex. While Wilson never uses any sarcasm in his essay to portray what he wants the readers to know and understand. Wilson gives logical and understandable reasons which can easily be seen with no rebuttal. In conclusion this is why I firmly believe that Wilson has a more coherent and cohesive, well stated, argument than Judy?s by no stretch of the imagination. Wilson?s thoughtful process in coming up with an essay that was factual, organizational, and had good use of language is why this argument is so well written. Judy?s essay is undeniably poorly written, to an extent to which a child may right. All in all, ?Just Take Away Their Guns? Wilson has captured the attention of the audience that is unmatched in Judy?s. So all I say is that, ?Guns do kill, people don?t?.

The Existence Of Aliens :: social issues

The Existence Of Aliens Do you believe in Aliens or life on different planets? There are many people who believe they seen alien or U.F.O sightings, abductions, crashes and animal mutilations But is this all true? There have been many U.F.O sightings around the world one sighting was on June 24th 1947 Kenneth Arnold was flying over the Cascade Mountains when a set of nine strange looking objects flying across the sky. At first he thought they were reflections of the sun but after looking at them more closely he realized that they were objects flying in formation. The objects he saw had wings what were curved and shaped like a crescent moon. His next thought was that they were some new aircraft that the Air Force was developing and he started to time them on his clock. He calculated that they were flying much faster than any aircraft he had read about. This was a strange case indeed was this alien space ships or was the government making aircrafts if so why where they never reviled. One of the first people to claim to have been abducted by Aliens where Betty and Barney Hill. During September 1961 they were driving back from a holiday in Canada through the White Mountains. They noticed a light that seemed to be following them they assumed that it was an airplane or helicopter. After driving for a while they were forced to stop the car because the light had descended rapidly and was now much closer. Barney got out to get a better look with the aid of his binoculars, and described the object as banana like with pointed tips and windows. Barney started to walk slowly towards the craft, and it was only after several desperate calls from his wife's that he returns to the car. He claimed to have see aliens through the window. At this they drove back home. However, they soon realized that more than an hour of time was 'lost' and noticed that the car had strange blotches on it. Betty reported it to Pease Air Force Base who confirmed that they had indeed tracked an unknown object around that time and location. Soon afterward Betty began having strange nightmares involving 'aliens'. She consulted several doctors who suggested hypnosis. Dr Benjamin Simon who was amazed by what the couple were independently describing hypnotized both the Hills.

Young Goodman Brown - A Thematic Illustration :: essays research papers

Nathaniel Hawthorne comes from an interesting background. He was born in Salem and later returned to live there. He was a descendant of William Hathorne, a puritan judge who persecuted Quakers, and John Hathorne, a puritan magistrate who participated in the Salem witch trials. Hawthorne's kinship to these two notables of puritan history makes the story "Young Goodman Brown," all the more interesting. Hawthorne alludes to John Hathorne when he writes about Goodman Brown's "fellow traveler" commenting on Brown's grandfather, who "lashed the Quaker woman so smartly through the streets of Salem." "Young Goodman Brown" is about one man's journey through the woods with the devil and his encounters that make him doubt his faith in himself, his wife, and the community in which they reside. The theme of this story is that beyond any intangible evil, the evil that men do is ultimately the more damaging. Throughout the story Hawthorne uses setting and characters as symbols representing different aspects of good and evil and he uses the plot to develop the eventual win-over of evil over "Goodman" Brown's "Faith." Not surprisingly "Young Goodman Brown" takes place in Salem during the puritan era. The story begins with Goodman Brown departing from his wife in the village to meet with and take a stroll in the forest with a "fellow-traveler" the devil. The contrast between the forest and the town is symbolic. On the outside, it seems like a normal, religious puritan village, but when one goes in deep, one sees there is a center of darkness. The deep, dark forest in the puritan town represents the internal evil of the villagers. The forest is viewed as mysterious, unknown and inhabited by the devil, while the town is pleasant safe and where his wife, "Faith," is. During Goodman Brown's walk through the "dark forest," he sees and learns that many of his mentors and relatives have chosen the path of evil. The forest is where all the respectable people of the town go to vent their evil while outside of the forest, they seem like they are pure and good. Hawthorne adds to the symbolism by personifying the trees "which barely stood aside to let the narrow path creep through" as Brown "walks alongside a dreary road." Hawthorne uses the characters of the story also to represent good and evil. The names of the main character and his wife are ironic. Faith, in the literal context of the story, is Goodman Brown's wife.

Allegory of the Cave Summary and Response Essay

Marlo Diorio Dr. Mishra – College Writing I â€Å"Allegory of the Cave† â€Å"Allegory of the Cave†, written by Plato, is story that contrasts the differences between what is real and what is perceived. He opens with Glaucon talking to Socrates. He has Glaucon imagine what it would be like to be chained down in a cave, not able to see anything other than what is in front of him. He tells a story of men that were trapped in a cave and were prisoners to the truth. These prisoners have only seen shadows. But because of their ignorance, these slaves to the cave believe that the shadows are real. The story goes on to say that one of the men has been dragged out of the cave. He is not happy to see the real world, yet upset because he is being taken away from all that he knows. As he approaches the outside, he is blinded by the sunlight that he has never seen. The sunlight can be interpreted as actual sun or as knowledge, making the journey rather painful in mental and physical ways. The prisoner wants to return to his life as a peasant inside of the cave. When he is outside of the cave, he only wants to look at shadows and reflections, but later proceeds to look at actual objects surrounding him. Lastly, he looks at the sun itself, as he realizes that is what created this beautiful nature. The climax of the story is when the ma realized that he no longer has to worry about reality and reasoning, because he achieved the understanding of it. Eventually, he goes back to the cave. He is not greeted nicely back at the cave because he is seen to have taken a meaningless trip. The man who had seen the outside world took it upon himself to teach the others and lead them into understanding the truth of reality. The story finished by Socrates saying that the most qualified and wise people are the best options for leading in government, like the prisoner who discovered truth. Throughout the reading of this symbolic narrative, I found it easy to relate to my own life. This story can be taken as an extended metaphor with a great meaning. I took this story as, the hard path to take out of the cave, was like the pattern of life. It would never be an easy path to walk down, and it would take a lot of struggling. Only certain determined people will actually make it to the opposite side. Socrates says these most qualified people should be the ones to lead the public. I believe this is also true in today’s society. I say this because when it comes to election time, we as a country are not going to vote for an uneducated lunatic. I believe that the president should be someone intelligent with good morals and very qualified. In order to reach that high point, you must go out of your comfort zone, like the prisoner did. In life, people go out of their comfort zones all of the time. I’ve always believed that in order to achieve something you’ve never had/done, you must do something you’ve never done before, such as stepping out of your comfort zone. Only the best can be found when you make an attempt to extend yourself as a human being. I relate the cave in this story to the social norm. No one wants to step out of it because I their life, the norm is all there is. I believe the shadows would represent all of the other things that could be out there, but they have no desire to go find out what they are. They are too comfortable with what they have and haven’t gone looking for more. The cave is a comfort zone for the prisoners in Plato’s time and for teenagers today. Without the outside world, there is no curiosity, no questioning. I believe it is important to question everything, because with questions come answers. And when you find out something you didn’t know, you’re learning. Yes, ignorance is bliss, but ignorance won’t make you president.

Design of Water-Based Fire Protection Systems Essay

Storage Tanks are the tanks in which flammable and combustible liquids are stored. The storage tanks should be constructed of steel in case they are constructed above grounds. The non-combustible substance can be used for the construction if it is used to store a combustible liquid. The material used for the construction must be that helps the tank to be protected against the fire. In one of the storage tank fires broke out at the island of Palau Merimau, Singapore and the fire started catching tankage containing petroleum, motor gasoline and diesel product. Despite the size of fire and adverse logistics, fire fighting efforts were successful in controlling the fire. This require operational and design engineering considerations, proper fire fighting strategy, prefire and emergency response plans, fire water management and fixed/semifixed system. A permanent water supply, of sufficient volume, duration, pressure, required to properly operate the fire fighting equipment shall be made available as soon as combustible materials accumulate and in the same way underground waters are to be provided, installed and made available for use as early as possible at the place of fire. In case were fixed water protection system is not installed with the tank with flammable or combustible liquids the floating roof to cover on liquid which helps to protect the fire to be broke out. Most fires in this design of tank burn only at the seal and are usually easily extinguished. Tanks with floating roofs are not likely to be involved in serious fires. This is because there is far less liquid surface is exposed to the fire. When storage tanks are not equipped with fixed fire protection then it must contain a stable liquid storage at a pressure 17kPa (gauge). The tanks containing flammable liquids should be normally kept closed for protection of unless the tank is venting. Where drainage facilities are provided to drain water from dike areas, efforts should be made to prevent flammable liquids from entering into any natural resources as it constitute fire hazard.
